In the neighbourhood of? A few sickles, say, five? Seven? I'm terrible at estimations...I don't think it would be too much.
I don't think the Knight Bus would charge fare based on distance - remember, the bus can teleport anywhere on land, so you can go from the Isle of Wight to the Outer Hebrides in seconds.
Therefore, I think the fare is simply a base fee (let's say ten sickles), plus some extra for where you want to go. The Knight Bus seems to go to destinations based on alphabetical order (i.e. Aberdeen to Anglesey), so additional fare could be based on the distance between destinations judging by their first letter. But with that said, the Ministry of Magic and the Leaky Cauldron are just one letter apart, so it shouldn't cost too much.
That's just what I think.

Actually, the Ministry of Magic and the Leaky Cauldron are as far apart as possible, because after the Ministry of Magic (M), you would have to go through the rest of the alphabet (N, O, P... till Z), and then from A till L (A, B, C, D... till L) so I doubt it'll be cheap then.
I think they would have a certain order of places they went. You have to remember that they generally only go somewhere if:
a) a person sticks out their wand
or
b) they are dropping the person off there.
So, if no one else was on the bus/getting on the bus, then there would be no need to stop anywhere else, which means they could pretty much just go where they needed to go.
